Grapevine berry colour mutants: molecular identification and phenolic composition
Abstract
Portugal, one of the Mediterranean countries with highest density of autochthonous cultivars, has 169 red, 152 white, and 22 grey cultivars approved for wine production (Ordinance 380/2012, 22nd November). Within the 343 different cultivars, there are some designations only varying for berry colour as Moscatel Galego Branco and Moscatel Galego Roxo. However, should they all be considered berry colour somatic mutants? In this study, nine cultivars (three Pinot used as reference, two Pique-Poul, two Malvasia Rei and two Carrega) varying in the berry skin colour were analysed using twelve microsatellite loci in order to correctly designate the original variety. Besides the three Pinot cultivars also Pique-Poul Gris and Pique-Poul Noir were confirmed as mutants revealing the same alleles in all the microsatellite loci analysed.
Phenolic compounds detection and quantification in the three Pinot and two Pique-Poul cultivars, comprising the non-coloured phenolics total concentration, was higher in Pinot Gris and lower in Pique-Poul Noir. On the other hand, anthocyanins total concentration was higher in Pinot Noir and much lower in Pique-Poul Gris. Two coumarilated anthocyanins were found in relatively large amounts in Pique-Poul Gris but were not detected in Pinot Gris.
