D-wines: use of LC-MS metabolomic space to discriminate italian mono-varietal red wines

Abstract

Studying wine metabolome through multiple targeted methods is complicated and limitative; since grapes, yeasts, bacteria, oxygen, enological techniques and wine aging collaborate to deliver one of the richest metabolomic fingerprint. Therefore, untargeted metabolomics, that developed and evolved as a consequence of the need to obtain a comprehensive characterization of the organic molecules in any biological sample, is the current methodology offering the best coverage of wine metabolome. Taking into account the large genetic diversity, the diversity of the climate and of the agronomical practices, and the wide winemaking culture characterizing the Italian wines, the metabolomic untargeted approach appears as an appropriate analytical tool to study such metabolic space. 

According to the national project D-Wines, 110 single-cultivar red wines from the 2016 vintage were collected directly from wineries across different regions of Italy: Sangiovese from Tuscany and Romagna, Nebbiolo from Piemont, Aglianico from Campania, Nerello Mascalese from Sicily, Primitivo from Apulia, Raboso and Corvina from Veneto, Cannonau from Sardinia, Teroldego from Trentino, Sagrantino from Umbria, and Montepulciano from Abruzzo. The wines were analyzed according to a well-defined RP-UPLC-HRMS-QTOF-MS protocol. 

The results of the data analysis, after their validation: a) confirmed untargeted LC-MS-based metabolomics as a powerful authenticity tool; b) provided indications about the similarity between the cultivars, clustering the wines in three major groups (Primitivo – Nebbiolo, Corvina, Raboso, Sangiovese – Teroldego, Sagrantino, Cannonau, Nerello, Aglianico, Montepulciano); c) furnished a rich list of putative markers characterizing each cultivar, where Primitivo, Teroldego and Nebbiolo had the maximum number of unique putative markers; d) revealed that the putative markers were not only phenolic metabolites; and e) pointed out rt/mz chromatographic sections helpful to distinguish each cultivar from the others. 

This study, together with other D-Wines analytical results, is directed to understand the diversity of Italian red wines and to characterize them in term of metabolic space coverage/variability and taste and in consequence comprehend better their quality.

Unravel the underlying mechanisms of delaying ripening techniques in grapevine

In a scenario of changing climate conditions, grapevine is significantly affected at multiple levels. Advancements in phenology and berry ripening, however, are the major dynamics of the generalized increase in average temperature and evaporative demand, negatively affecting berry quality and productivity. The aim of this work was to unravel the underlying mechanisms of bunch-zone auxin application (NAA; 1-Naphthaleneacetic acid) and source-limiting canopy management approaches in delaying berry ripening. In randomized block design experiments, control vines were compared to vines treated with NAA, subjected to apical-to-bunch defoliation or antitranspirant application (n=10-to-42 plants per treatment).

Effectiveness of carboxymethyl cellulose (CMC) on tartaric stabilization of cava base wine

Recent EU regulations allow the use of carboxymethylcellulose (CMC) as a stabilization agent in wine. We tested CMC in bases for sparkling wines, which must be stabilized before the second fermentation that raises alcohol concentration by 1,5%.

The challenge of quality in sulphur dioxide free wines: natural polyphenol alternatives

Sulphur dioxide (SO2) seems indispensable in winemaking because of its properties. However, a current increasing concern about its allergies effects in food product has addressed the international research efforts on its replacement. This supposes a sufficient knowledge of its properties and conditions of use. Several studies compared SO2 properties against new alternatives that are supposed to overcome SO2 disadvantages. Firstly, the state of art on SO2 wine replacements is revised, and secondly, the last promising results using natural enriched polyphenol extracts are shown.

New acylated flavonols identified in the grape skin of Vitis vinifera cv. Tannat and their wines

Flavonols are a class of flavonoid compounds derived from plant secondary metabolism. There they play different roles like antioxidants, internal regulators and UV screenings. In red wines, flavonols have increasingly received consideration by part of scientific and winemakers according their properties began to arise known. Among these stand out wine colour stabilization and their value as bioactive compounds. In this work the complete series of the acetylated and p-coumaroylated derivatives of the 3-O-glycosides of methoxylated flavonols, namely isorhamnetin, laricitrin and syringetin, have been identified in grapes and their respective wines from Vitis vinifera cv. Tannat.

Results of late-wurmian to present-day climatic-geological evolution on to spatial variability of pedologic-geological characters of the AOC Gaillac terroirs (Tarn, Midi-Pyrénées)

The AOC Gaillac area is divided into three main terroirs : « The left bank terraces », « The right bank coteaux » and
« The plateau Cordais ». This division is valid at a regional scale, but it suffers of a number of local-scale exceptions. This spatial variability of the pedologic-geologic characteristics at the plot scale has been derived mainly from the main late-Würmian solifluxion phase occurring at the transition between the peri-glacial climate and the Holocene temperate conditions (13,000-10,000 yrs BP).
