Detoxification capacities of heavy metals and pesticides by yeasts 

Abstract

Winegrowing is still characterized by the extensive use of chemical fertilizers and plant protection products, despite strong recommendations to limit these practices. A part of these xenobiotics and metals are then found in grape juice and wine, causing a major health concern, as well as negatively affecting the fermentation process. In recent years, there has been renewed interest in non-Saccharomyces yeasts. These species have a wide phenotypic diversity, which would be exploited to broaden the aromatic palette of wines. The potential of some of these species as a bio-protection agent, in pre-fermentation treatment, has also been established.

In addition to these characteristics, the heavy metal sorption capacities of these species are mentioned in the literature. Pre-fermentation treatment of the harvest with non-Saccharomyces (NS) yeasts therefore appears to be an alternative to the use of chemical inputs for the elimination of organic and inorganic xenobiotics from grape musts. However, our knowledge of the detoxification capacities of yeasts and their molecular and biochemical determinant is not yet sufficiently advanced to allow such methods to be developed and put into practice.  In this study, we investigated the detoxifying abilities of fourteen strains of Saccharomyces and non-Saccharomyces yeasts during the first stages of wine fermentation. Fermentations were conducted in synthetic must in the presence or absence of pesticides (mix of twenty pesticides commonly used in the vineyard) / metals (copper). The colorimetric determination of copper concentration throughout fermentation revealed that most Saccharomyces and non-Saccharomyces yeasts detoxify almost all copper in less than 48 hours. Regarding pesticides detoxification, quantified by GC-MS (Dubernet laboratory), a higher variability was observed, with a removal of 0% to 90% of the compound depending on the yeast strain and the pesticide. In conclusion, this study showed the great diversity of the capacity of yeasts to detoxify molecules used in vineyards to fight against diseases and/or parasites and the biotechnological potential that this represents.

Molecularly imprinted polymers: an innovative strategy for harvesting polyphenoles from grape seed extracts

Multiple sclerosis (MS) is a multifactorial autoimmune disease associating demyelination and axonal degeneration developing in young adults and affecting 2–3 million people worldwide. Plant polyphenols endowed with many therapeutic benefits associated with anti-inflammatory and antioxidant properties represent highly interesting new potential therapeutic strategies. We recently showed the safety and high efficiency of grape seed extract (GSE), a complex mixture of polyphenolics compounds comprising notably flavonoids and proanthocyanidins, in an experimental autoimmune encephalomyelitis (EAE) mouse model of MS.

Dynamics of Saccharomyces cerevisiae population in spontaneous fermentations from Granxa D’Outeiro terroir (DOP Ribeiro, NW Spain)

Granxa D’Outeiro is a recovered ancient vineyard located in the heart of DOP Ribeiro, where traditional white grapevine varieties are growing under sustainable management. Spontaneous fermentations using grape must from Treixadura, Albariño, Lado, Godello, and Loureira varieties were carried out at experimental winery of Evega. Yeasts were isolated from must and at different stages of fermentation. Those colonies belonging to Saccharomyces cerevisiae were characterized at strain level by mDNA-RFLPs.

Association between dietary pattern and wine consumption and Alzheimer’s disease in a cohort from La Rioja (Spain)

Addressing modifiable risk factors is the most promising strategy to prevent/delay Alzheimer Disease (AD)[1]. The aim of the study was to establish the connections between dietetic habits, wine consumption and AD. Thus, 98 volunteers were recruited: 50 diagnosed as AD and 48 healthy/controls. The Food Frequency Questionnaire (FFQ) was used for dietary patterns assessment and, based on these data, the Mind Diet Score was calculated. (Poly)phenol metabolites (especially derived from wine consumption) were analyzed by UPLC-QqQ-MS/MS in 24-h urine samples to confirm dietary (poly)phenol consumption.

Lipids at the crossroads of protection: lipid signalling in grapevine defence mechanisms

Understanding grapevine molecular processes and the underlying defence responses is vital for developing sustainable disease control strategies. Lipid signalling pathways, involving the synthesis and degradation of lipid molecules, have emerged as a key regulator in plant defence against pathogens. This study aims to elucidate the role of fatty acids and lipid signalling in grapevine’s defence response to P. viticola infection. The expression of lipid metabolism-related as well as lipid signalling genes was analysed, by qPCR, in three grapevine genotypes: Chardonnay (susceptible), Regent (tolerant) with Rpv3-1 resistance loci, and Sauvignac (resistant) harbouring a pyramid of Rpv12 and Rpv3-1 resistance loci.

Drought tolerance assessment and differentiation of grapevine cultivars using physiological metrics: insights from field studies

This study aimed to validate a protocol and compare metrics for evaluating drought tolerance in two Vitis vinifera grapevine cultivars under field conditions. Various metrics were calculated to represent the physiological responses of plants to progressive water deficit. Data were collected from Sauvignon Blanc and Chardonnay plants subjected to three irrigation levels during the 2022-2023 season, along with data from three previous seasons. Hydro-escape areas were used to assess the plant’s ability to reduce water potential with decreasing soil water availability.
