Protection of genetic diversity: maintenance and developements of a grapevine genebank in Hungary

Abstract

Among the items preserved in gene banks, the old standard and autochthonous varieties represent an increasing value, since these varieties may have properties to make their cultivation more effective under changing climatic conditions. The increasingly extreme weather is a huge challenge for the viticulture. Collectional varieties can also play important role in protection against pests and pathogens. A genebank ensures not only the preservation of rare varieties, but also gives the opportunity for more knowledge and research of these varieties.
Our main goal is to further develop national grape database, namely collecting and managing ampelographic and ampelometric descriptive data (OIV descriptors), imaging files, providing information on the origin of the varieties based on the data related to their pedigree, including parents, family trees and offspring.
We examine all 1.570 items in our gene bank with special attention to about 117 grape varieties autochthonous to the Carpathian Basin. From this collection more than 50 varieties can neither be found in national genebanks, nor in larger international collections, such as Bakhtiori Chernyi, Balsare Blanc, Cornucopia, Jabizlak, Piros Gránát, Kárpáti Rizling, Krabljak, Sesh i Zi, Polombina, or Tuingirni kara. Some of them are native to the Carpathian Basin, while others are mostly oriental origin. The genebank contains individuals propagated from the methuselah vine stock of Europe, such as the 450-500-year-old Rosa Menna di Vacca in city of Pécs (validated by SSR markers). Our research is also enriched by the Zametovka variety growing in Maribor (Slovenia), which considered as the oldest vine in Europe.
As a result of our work, the database system will also provide data obtained by molecular methods (SSR, SNP), which identify the genotypes supporting for the discovery of further relationships and for further research about the origin of the domesticated grapevine.
The most important international and autochthonous varieties of the genebank will be analyzed on berry skin and flesh anthocyanin composition as well as aroma profiles.

Winemaking processes discrimination by using qNMR metabolomics

AIM: Metabolomics in food science has been increasingly used over the last twenty years. Among the tools used for wine, qNMR has emerged as a powerful tool to discern wines based on environmental factors such as geographical origin, grape variety and vintage (Gougeon et al., 2019a).

Grapevine varietal diversity as mitigation tool for climate change: Agronomic and oenologic potential of 14 foreign varieties grown in Languedoc region (France)

Climate change effects in Languedoc include an expected rise in temperatures, increased evapotranspiration as well as more severe and frequent climatic hazards, such as frost, drought periods and heat waves. For winegrowers theses phenomena impact both yield and quality, resulting in more frequent unbalanced wines. Research on identified mitigation tools for vineyard management is necessary to improve resilience of grapevine agrosystems. Varietal assortment is one of them. This study focuses on agronomic and oenologic potential of 14 foreign varieties grown in Languedoc French region. Fourteen grapevine varieties were monitored during 2021 from June until harvest on eight different sites, some of which occurring on more than one site adding up to 21 different modalities: 7 white varieties Alvarinho B, Assyrtiko B (2), Malvasia Istriana B, Parellada B, Verdejo B, Verdelho B, Xarello B, and 7 black varieties Saperavi N (2), Touriga nacional N, Baga N, Aleatico N, Montepulciano N (2), Primitivo N (3), Calabrese N (3). Varietals were compared through the following parameters: phenology was assessed by using the information collected in the Database Network of French Vine Conservatories (INRAE-SupAgro-IFV, 2005-2015). The number of inflorescences for shoots from secondary buds and bourillons and suckers were observed to assess post-bud break frost tolerance potential. Grapevine water status was studied through stem water potential measurement, observation of foliage symptoms of drought, and 𝛿13C on must. Frequencies and intensities of downy mildew, powdery mildew, and black rot attacks were estimated before harvest on leaves and clusters and botrytis at harvest to assess disease susceptibilities. Berry composition was monitored from end of veraison until harvest. Yield and mean bunch weight were also calculated. Varieties were then ranked on a 1-4 scale for each parameter and compared through PCA. Forty two stations of the Mediterranean basin were compared by PCA with the Multicriteria Climatic Classification indicators in order to confront the collected information during 2021 campaign to the hypothesis that plants coming from dry and hot regions are genetically adapted to such climatic conditions.

Measurement of synthetic solutions imitating alcoholic fermentation by dielectric spectroscopy

Having the possibility to use a wide spectrum of elecromagnetic waves, dielectric spectroscopy is a technique commonly used for electrical characterization of dielectrics or that of materials with high energy storage capacity, just to name a few. Based on the electrical excitation of dipoles (polymer chains or molecules) or ions in relation to the characteristics of a weak external electric field, this method allows the measurement of the complex permittivity or impedance of polarizable materials, each component having a characteristic dipole moment.In recent years, the food industry has also benefited from the potential offered by this technique, whether for the evaluation of fruit quality or during the pasteurization of apple juice [1-3]. As the tests are fast and do not destroy the products, dielectric spectroscopy proved to be an experimental tool suitable for online measurements as well as long-term monitoring.

Optimization of in vitro establishment of grapevine varieties for fast micropropagation 

Micropropagation is an important alternative to conventional methods of plant propagation. The objective of this study was to optimize a protocol for in vitro micropropagation of selected grapevine hybrids (H19 and H20) that are included in our breeding program. For the sprouting initiation experiment, nodal cuttings with only one axillary bud from two hybrids were separated, disinfected, and cultivated in 50% Murashige Skoog nutrient medium (½ MS) and Woody Plant Medium (WPM), adding 4.4 µM benzyladenine (BA) in both mediums.
