Generation of radicals in wine by cavitation and study of their interaction with metals, phenols and carboxylic acids

Abstract

High-power ultrasounds have been related to an accelerated aging of wines, an effect that has been associated to the formation of radical species caused by the cavitation phenomenon [1]. This phenomenon consists of the formation of bubbles in the liquid medium that, when they collapse, cause high-pressure hot spots and temperatures of up to 4800 k [2], notably increasing the reactivity in the medium. Although until now it has been considered that cavitation causes the homolytic rupture of the water molecule, generating hydrogen and hydroxyl radicals [3], it must be taken into consideration that the presence of these radicals may have other origins, such as the reaction of fenton: the main source of hydroxyl radicals in biological systems (eq. 1). This reaction could be induced in wines by the presence of fe (ii) and hydrogen peroxide, which can be generated by the reduction of diatomic oxygen thanks to the presence of phenolic compounds, antioxidants, in wine [4]. Fe+2 + h2o2→ho∙+oh- + fe+3 (eq. 1) until now, it has not been possible to reliably demonstrate the formation of hydroxyl radicals due to the use of ultrasound, nor its effect on some main components of wine: ethanol, carboxylic acids, phenols and metals, such as fe (ii). Furthermore, although the generation of hydroxyl radicals could be linked to the cavitation phenomenon, their intensification and reactivity can be influenced by the different components of the wine, and the participation of the fenton reaction in the process is even possible. Thus, the objective of this work was to demonstrate the generation of radicals due to the phenomenon of cavitation induced by ultrasound and to verify if the fenton reaction also participates in their formation, for which a real wine and a model wine with ethanol, acids carboxylic acids, phenols and metals were used. The wines were subjected to the application of high-power ultrasound and the generation of radicals and their interaction with the components present in the medium were determined. Furthermore, the hydrogen peroxide content was analyzed as evidence of the participation of the fenton reaction in the formation of radicals. The determination of the radicals present in the wines was carried out using the electron spin paramagnetic resonance (epr) spectroscopy technique, using 5,5-dimethyl-1-pyrrolin-n-oxide (dmpo) as a “spin-trap”. Furthermore, regarding its interaction with other components, the oxidation of caffeic acid and catechin was determined by reverse phase hplc. The results demonstrated the generation of ho· and h· radicals by the application of ultrasound (60 w, 30 khz), with no hydrogen peroxide detected, which indicated that the fenton reaction had not occurred. These unstable species reacted completely with ethanol (12%) generating 1-hydroxyethyl radicals. They also generated carbon-centered radicals with tartaric acid and phenolic compounds (caffeic acid and catechin). However, the amount of radicals was reduced by the presence of fe (ii) and its oxidation to fe (iii). It was also observed that metals promote the stabilization of semiquinones in the presence of oxygen, which agrees with what has been described in the literature on the formation of an equilibrium of valence tautomers [5]. Furthermore, the preference of oxygen for the radicals generated favored the formation of non-radical products.

Generación de radicales por cavitación en vino y estudio de su interacción con metales, fenoles y ácidos carboxílicos

Los ultrasonidos de alta potencia han sido relacionados con el envejecimiento acelerado de los vinos, efecto que se ha asociado a la formación de especies radicalarias originadas por el fenómeno de cavitación [1]. Este fenómeno consiste en la formación de burbujas en el medio líquido que, al colapsar, provocan puntos calientes de alta presión y temperaturas de hasta 4800 k [2], aumentando notablemente la reactividad en el medio.  Si bien hasta ahora se ha considerado que la cavitación provoca la ruptura homolítica de la molécula de agua, generando radicales hidrógenos e hidroxilos [3], ha de tenerse en consideración que la presencia de estos radicales puede tener otros orígenes, como es la reacción de fenton: la principal fuente de radicales hidroxilos en sistemas biológicos (ec. 1). Esta reacción podría ser inducida en vinos por la presencia de hierro (ii) y peróxido de hidrógeno, que puede generarse por la reducción del oxígeno diatómico gracias a la presencia de los compuestos fenólicos, antioxidantes, en vino [4]. Fe+2 + h2o2→ho∙+oh- + fe+3  (ec. 1) hasta ahora no se ha podido demostrar fehacientemente la formación de radicales hidroxilos debido al uso de ultrasonidos, así como tampoco su efecto sobre algunos componentes principales del vino: etanol, ácidos carboxílicos, fenoles y metales, como fe (ii). Además, si bien la generación de radicales hidroxilos pudiera vincularse al fenómeno de cavitación, su intensificación y la reactividad de los mismos puede verse influenciada por los distintos componentes del vino, siendo posible incluso la participación de la reacción fenton en el proceso. Así, el objetivo de este trabajo fue demostrar la generación de radicales por el fenómeno de cavitación inducida por ultrasonidos y verificar si en su formación también participa la reacción de fenton, para lo que se emplearon un vino real y un vino modelo con etanol, ácidos carboxílicos, fenoles y metales. Los vinos fueron sometidos a la aplicación de ultrasonidos de alta potencia y se determinó la generación de radicales y su interacción con los componentes presentes en el medio. Además, se analizó el contenido de peróxido de hidrógeno como evidencia de la participación de la reacción fenton en la formación de radicales. La determinación de los radicales presentes en los vinos se realizó mediante la técnica de espectroscopía de resonancia paramagnética de espín electrónico (epr), utilizando 5,5-dimetil-1-pyrrolin-n-oxide (dmpo) como “spin-trap”. Además, en cuanto a su interacción con otros componentes, se determinó la oxidación de ácido cafeico y catequina mediante hplc en fase reversa. Los resultados demostraron la generación de radicales ho· y h· por la aplicación de ultrasonidos (60 w, 30 khz), no habiéndose detectado peróxido de hidrógeno, lo cual indicó que no había ocurrido la reacción fenton. Estas especies inestables reaccionaron completamente con el etanol (12%) generando radicales 1-hidroxietilo. También generaron radicales centrados en carbono con el ácido tartárico y compuestos fenólicos (ácido cafeico y catequina). No obstante, la cantidad de radicales se vio reducida por la presencia de fe (ii) y su oxidación a fe (iii). También se observó que los metales promueven la estabilización de semiquinonas en presencia de oxígeno, lo que concuerda con lo descrito en literatura sobre la formación de un equilibrio de tautomerismos de valencia [5]. Además, la preferencia del oxígeno por los radicales generados favoreció la formación de productos no radicalarios.

Génération de radicaux par cavitation dans le vin et étude de son interaction avec les métaux, phénols et acides carboxyliques

Les ultrasons de haute puissance ont été associés au vieillissement accéléré des vins, effet associé à la formation d’espèces radicalaires provoquées par le phénomène de cavitation [1]. Ce phénomène consiste à la formation de bulles dans le milieu liquide qui, lorsqu’elles s’effondrent, provoquent des points chauds à haute pression et des températures pouvant atteindre 4 800 k [2], augmentant notamment la réactivité du milieu. Bien que jusqu’à présent on ait considéré que la cavitation provoque la rupture homolytique de la molécule d’eau, générant des radicaux hydrogène et hydroxyle [3], il faut tenir compte du fait que la présence de ces radicaux peut avoir d’autres origines, comme la réaction de fenton: la principale source de radicaux hydroxyles dans les systèmes biologiques (éq. 1). Cette réaction pourrait être induite dans les vins par la présence de fer (ii) et de peroxyde d’hydrogène, qui peut être généré par la réduction de l’oxygène diatomique grâce à la présence de composés phénoliques, antioxydants, dans le vin [4]. Fe+2 + h2o2→ho∙+oh- + fe+3 (éq. 1) jusqu’à présent, il n’a pas été possible de démontrer de manière fiable la formation de radicaux hydroxyles due à l’utilisation des ultrasons, ni son effet sur certains composants principaux du vin: l’éthanol, les acides carboxyliques, les phénols et les métaux, comme le fe (ii). De plus, bien que la génération de radicaux hydroxyles puisse être liée au phénomène de cavitation, leur intensification et leur réactivité peuvent être influencées par les différents composants du vin, et la participation de la réaction de fenton au processus est même possible. Ainsi, l’objectif de ce travail était de démontrer la génération de radicaux due au phénomène de cavitation induit par les ultrasons et de vérifier si la réaction de fenton participe également à leur formation, pour laquelle un vin réel et un vin modèle avec de l’éthanol, des acides carboxyliques, phénols et métaux ont été utilisés. Les vins ont été soumis à l’application d’ultrasons de haute puissance et la génération de radicaux ainsi que leurs interactions avec les composants présents dans le milieu ont été déterminées. Egalement, la présence de peroxyde d’hydrogène a été analysée comme preuve de la participation de la réaction de fenton à la formation de radicaux. La détermination des radicaux présents dans les vins a été réalisée par la technique de spectroscopie par résonance paramagnétique électronique de spin (epr), en utilisant le 5,5-diméthyl-1-pyrrolin-n-oxyde (dmpo) comme « spin-trap ». En plus, en ce qui concerne son interaction avec d’autres composants, l’oxydation de l’acide caféique et de la catéchine a été déterminée par hplc en phase inverse. Les résultats ont démontré la génération de radicaux ho· et h· par l’application d’ultrasons (60 w, 30 khz), sans détection de peroxyde d’hydrogène, ce qui indique que la réaction de fenton ne s’est pas produite. Ces espèces instables ont réagi complètement avec l’éthanol (12 %) en générant des radicaux 1-hydroxyéthyle. Ils ont également généré des radicaux centrés sur le carbone avec l’acide tartrique et des composés phénoliques (acide caféique et catéchine). Cependant, la quantité de radicaux a été réduite par la présence de fe (ii) et son oxydation en fe (iii). Il a également été observé que les métaux favorisent la stabilisation des semiquinones en présence d’oxygène, ce qui est en accord avec ce qui a été décrit dans la littérature sur la formation d’un équilibre de tautomères de valence [5]. De plus, la préférence de l’oxygène pour les radicaux générés favorisait la formation de produits non radicalaires.

To characterize a wine, the most frequently used criteria describe its color, its origin, the grape varieties from which they come, or even for white wines its residual sugar content (dry, semi-dry, sweet). In france, the system of appellations of origin set up in 1919 was initially based solely on the notoriety and origin of the wines. But given the unfavorable consequences that this lack of details generated, the public authorities quickly integrated in 1927 into the “capus” law criteria for access to designations of origin, relating to the specific characteristics of the soils of the vineyards and the grape varieties used, in particular exclusion of interspecific hybrid varieties. In 1935 the creation of the aoc system confirmed the interest in precisely defining all the production conditions that must be implemented to be able to claim the benefit of an aoc, and grape varieties were an essential condition for acquisition.
