Epigenetic Modulation Of Inflammation And Synaptic Plasticity By Polyphenolic Metabolites Promotes Resilience Against Stress In Mice

Abstract

Introduction: Major depressive disorder is associated with abnormalities in the brain and the immune system. Chronic stress in animals showed that epigenetic and inflammatory mechanisms play important roles in mediating resilience and susceptibility to depression.

Material & Methods: Here, through a high- throughput screening, we identify two phytochemicals, dihydrocaffeic acid (DHCA) and malvidin-3′-O-glucoside (Mal-gluc) that are effective in promoting resilience against stress by modulating brain synaptic plasticity and peripheral inflammation. DHCA/Mal-gluc also significantly reduces depression-like phenotypes in a mouse model of increased systemic inflammation induced by transplantation of hematopoietic progenitor cells from stress- susceptible mice.
Results: DHCA reduces pro-inflammatory interleukin 6 (IL-6) generations by inhibiting DNA methylation at the CpG-rich IL-6 sequences introns 1 and 3, while Mal-gluc modulates synaptic plasticity by increasing histone acetylation of the regulatory sequences of the Rac1 gene.

Conclusions:

This study suggests that polyphenolic metabolites capable of influencing peripheral inflammation and synaptic maladaptation may eventually be implemented as novel therapeutic interventions for promotion of mental health.

Report on the work carried out by the zoning group of the O.I.V.

La création officielle du groupe Experts Zonage Vitivinicole à l’O.I.V., qui s’inscrit dans la Commission Viticulture, est récente. Le Professeur Mario FREGONI en assure la présidence depuis 1998, assisté du vice-président et du secrétaire général Mario FALCETTI. Ils ont été confirmés dans leurs fonctions lors des sessions de mars 2001. Actuellement, le groupe d’experts Zonage Vitivinicole de l’O.I.V. se compose de 40 délégués, représentant 18 pays membres. La mise en place de ce groupe a tout d’abord été initiée par l’Instituto Agrario de San Michele (Italie) et l’Unité de Recherches Vigne et Vin du Centre INRA d’Angers (France). From soil to canopy, the diversity of adaptation strategies  to abiotic constraints in grapevine

Climate change is here. One of the main consequences is an increase in the frequency and severity of abiotic stresses which mostly occur in a combined manner. Grapevine, which grows in a large diversity of pedo-climatic conditions, has presumably evolved different mechanisms to allow this widespread adaptation. Harnessing the genetic diversity in these mechanisms will be central to the future of viticulture in many traditional wine growing areas. The interactions between the scion and the rootstock through grafting add an additional level of diversity and adaptive potential to explore.
At the physiological level, these mechanisms are related to processes such as root system development and functioning (water and nutrient uptake), interactions with the soil microbiome, gas exchange regulation, hydraulic properties along the soil-plant-atmosphere continuum, reserve storage, short and long distance signaling mechanisms and plasticity for some of these traits.
