Optimization and validation of a fully automated HS-SPME method for determination of VCCs and its application in wines submitted to accelerated ageing

Abstract

Wine aroma is a complex gaseous mixture composed of various compounds; some of these molecules derive directly from the grapes while most of them are released and synthetized during fermentation or are due to ageing reactions. Among the latter class of compounds, carbonyls are the principal products of oxidation reactions which take place during the storing time. Volatile carbonyl compounds (VCCs) are related to aromatic nuances of vanilla, caramel, butter, honey, potato, orange, lemon, violets, cider and plum, which are pleasant scents characteristics of oxidized wines. However, apart from cases where it is a deliberate process, oxidation is commonly undesired and the presence of a relevant content of carbonyls is related to aroma defects. Because of that, monitoring the concentration of VCCs could be added as a quality control for the evaluation of a complete fermentation, correct winemaking style, and proper bottling and storage. In this research an HS-SPME method1 was optimized and validated with the aim to be used as a tool to achieve this goal. The use of a solvent-free extraction allowed to maximize the coherence to the Green Analytical Chemistry principles with a simultaneous achievement in performance, reliability and robustness. In this method, all sample preparation steps were automated using the autosampler minimizing the human time consumption to enhance the scalability to routine analysis. As many as 46 VCCs (mainly linear aldehydes, Strecker aldehydes, unsaturated aldehydes, ketones, and many other) were the analytes under investigation. Drought affects vineyard soil microbiome: approach to select micro-organisms adapted to drought

Climate transition with frequent heat waves and long drought periods threatens grapevine productivity and wine quality in the Mediterranean regions. Microorganisms are known to contribute to plant fitness and to stimulate plant resilience against biotic and abiotic factors.
In this work, it was assessed the impact of long-term drought on soil microbiome associated to grapevine in open field in Alentejo, renowned Portuguese wine region.
Soil and plant tissues of drought tolerant Syrah cultivar exposed to three irrigation levels (100%- FI, 50%-DI ETc; rain-fed–NI) for 5 years were sampled for two years (2022-2023). Metabarcoding analysis of soil bacteria (16S V4 rRNA) and fungi (ITS sub-region) were integrated with soil physiochemical properties and leaves´ physiological data. Pre-dawn leaf water potential and stomatal conductance confirmed the imposed drought scenarios. Even though, α- and β-diversity of prokaryotic and eukaryotic microbial communities differed more by season than water availability, samples clustered according to soil water content and pH (p<0.05). Fungal communities show higher differences in the structure across treatments than bacteria. In 2023, 16 bacterial against 61 fungal ASVs were significatively different in abundance between NI and FI. Beijerinckiaceae, Bradyrhizobiaceae (Alphaproteobacteria) and Nocardioidaceae, Streptomycetaceae (Actinobacteria) families resulted to be significatively more abundant in NI, while Ascomycota, Basidyomicota and Mortierellomycota are the most important fungal phyla in NI. With culturomics data, this study aims to gather insights into how soil microbiome is remodelled under drought and contribute to select bacterial and fungal taxa with potential to mitigate drought stress in vineyards.

Infrared spectroscopy investigation of fresh grapevine organs for clustering and classification.

The spectral information acquired from fresh whole grapevine organs have yet to be fully explored. Infrared spectroscopy provides the means to rapidly measure fresh plant material and providing extensive information on the physical and chemical structure of samples.

Nolo alcohol beverages are produced to emulate the appearance, aroma, and taste of alcoholic beverages, potentially facilitating a sense of social integration when consuming a product that closely resembles alcohol.

May lactic acid bacteria play an important role in sparkling wine elaboration?

The elaboration of sparkling wine is a demanding process requiring technical as well as scientific skills. Uncovering the role of the terroir to the final product quality is of great importance for the wine market. Although the impact of the yeast strains and their metabolites on the final product quality is well documented, the action of bacteria still remains unknown. The malolactic fermentation (MLF) is carried out by the lactic acid bacteria after the alcoholic fermentation in order to ensure the microbial stability during the second fermentation that takes place in the bottle or in tanks. Oenococcus oeni is the only selected species to drive MLF that has been commercialized for sparkling wine elaboration and it is naturally present on grapes, in the cellar and also in the final product. However, whether the bacterial strain contributes to the sensory characteristics of sparkling wine is still questioned.

Multidisciplinary strategies for understanding ill-defined concepts

Aims: The objective of the present work is to review strategies applied to decrypt multidimensional and ill-defined concepts employed by winemakers and to illustrate these strategies with recent applications.

Effects Of Injections Of Large Amounts Of Air During Fermentation

Aim: Evaluating the effects of high amount of air injection during red wine fermentation process, on phenolic extraction dynamics, oxygen dissolution, phenolic compounds evolution, and oxidation of red wines.MethodsRed grapes musts were fermented in 100.000 L stainless steel tank, equipped with Parsec SRL “Air mixing” gas injection systems. For this experiment, treatments with two injection regimes, high and low intensity, and high and low daily frequency, were used. Oxygen analyzer was introduced into the tank to evaluate the gas concentration evolution along the fermentation.
